Nucleic acid purification and characterization scientist will lead efforts to purify and evaluate non-viral vector product performance and potency in relevant cell populations. Involves significant theoretical understanding of and practical experience with nucleic acid manufacturing process, with purification techniques, and with analytical methods used to characterize these kinds of molecules. Work to develop scalable and GMP-compliant processes required to purify different formats of nucleic acid-based therapeutic vectors. For the purification of plasmid DNA vectors produced by bacterial fermentation, develop appropriate lysis methods, cellular debris removal, and chromatography resin screening and buffer scouting to identify optimal binding and elution conditions to obtain highest purity plasmid vector or template products. For therapeutic mRNA vectors manufactured by in vitro transcription, candidate should have thorough understanding of RNA biochemistry and of enzymatic processes, including 5-end capping reactions. Develop, perform, and optimize purification strategies that effectively yield non immune-stimulatory, capped mRNA molecules. Strong analytics expertise is also required to thoroughly characterize nucleic acid vectors. Candidate will work collaboratively with cell biology and immunology colleagues to evaluate the potency and other critical quality attributes of different format nucleic acid vectors and will help develop and implement relevant high-throughput analytic assays. M.S. or B.S. degree with significant relevant experience in nucleic acid purification and characterization methods. English.
