Analyzing Synthesis

The University of Southern California’s Oscar Aparicio and his colleagues describe how they conduct a genome-wide analysis of DNA synthesis in this month’s CSHL Protocols. In their approach, they couple BrdU immunoprecipitation with DNA microarrays to find BrdU-labeled chromosomal DNA throughout the genome. “BrdU-IP-chip has many potential applications and has already been used to identify replication origins, make quantitative comparisons of origin firing between strains, and examine replication fork progression,” the authors write.
